I just did this fast video as a reply to another thread, but thought it would be interesting to share with the rest of you. It's my method to easily create palettes and color-separated sprites.

1. Convert the sprite into RGB mode
2. Select with the magic wand the areas that you want to recolorize. Add more selected areas by holding Shift key. Remove unwanted areas by holding Alt key.
3. Once you are happy with the selection, go to the Layers panel and click on "Create new fill or adjustment layer" and then select "Hue/Saturation"
4. In the new window, click on "Colorize" and then adjust the sliders until you get a desired color. ensure the "Preview" checkbox has been marked to see the changes in real time. Click "Ok" when done
5. Go to the layers pane, select the layer with the base sprite and repeat step 2 to select another area.
6. Once you are finished with all areas/colors, flatten all layers and convert the sprite back to Indexed mode.
7. You can reuse the same "Hue/Saturation" layers into other sprites by simply dragging the layers from one file to the other. To edit the mask on the new sprite, click Alt key on the mask to edit it.

Happy color separating!!
